There are three main techniques commonly used for microscopic observations: brightfield microscopy, darkfield microscopy and phase contrast microscopy. There is also a special technique, which uses polarized light for illumination, called polarized light microscopy. Polarization microscopy studies can be carried out using a simple brightfield microscope; the only additional accessory required is a polarizing filter.

Phase contrast microscopy requires the use of special optics (phase contrast objectives) and a special condenser (phase contrast condenser). Phase contrast microscopy is used when working with high magnifications of about 1000x for observing specimens that can’t be observed in detail using basic bright filed technique; for example, bacteria or living cells’ flagella and cilia. The cost of the special equipment required for phase contrast microscopy makes these microscopes a lot more expensive as compared to common optical microscopes. Because of their high price and relatively narrow range of applications, phase contrast microscopes are not favored by amateur users and are only available to a narrow circle of specialists.

This leaves us with the two most popular microscopic techniques – brightfield and darkfield microscopy. Isn’t it great to be able to enjoy the advantages of both these methods?
